(A) Localization of two mutants of Bcl-xL carrying single substitutions in the C-terminal α-helix, following their expression in yeast. The mutant A221R was excluded from the mitochondrial fraction, opposite to the mutant G217R.

(B) Localization of wild-type and A221R mutant of Bcl-xL in HeLa cells. Cells were transfected with Bcl-xL-expressing plasmids and Bcl-xL was detected by immunofluorescence as described previously [35]. Nuclei were counter-stained with DAPI. The mutant A221R displayed a diffuse cytosolic localization, opposite to the wild-type.
