SLAP-S25 targets both LPS and pEtN-LPS by competing with Mg2+ to dissect the tight joint between two neighboring lipids of LPS to increase the permeability of the outer membrane. pEtN-LPS is a modified LPS molecule. Importantly, SLAPS25 binds to the terminal group in the glycerol of PG, disrupting the integrity and function of the inner membrane. The collective behaviors of SLAP-S25 finally result in the intracellular accumulation of antibiotics to potentiate antibiotic efficacy.
